The only thing that possibly makes me think they're not actual sisters is that Akira's voice and even personality changes to be more squeaky, solid, and boy-like between September and November of last year. You can even notice she has a softer, and girlier voice when they debuted and during the videos around that time compared to her voice during their Channel Greeting vid published on Nov 23 2021

I don't know if maybe the mic they were using affected it, but Makoto's voice is still mostly the same but there's too much of a difference for Aki's voice that they might have quietly replace her with someone else after a certain point
