It has to be someone from Luxiem, they're the only ones where it would be a disaster for the company if anyone from that wave left before even 2 years. Not even the sisters would be able to explain it away, the branch's reputation would be permanently fucked at that point. And the only ones I can see ready to go are either Shu, Mysta, or Vox. Shu apparently gets into screaming matches with management behind the scenes and was devastated with AR Fes being cancelled, Mysta felt like he did all he wanted and would rather play gachas in peace and not think about management's 1% merch cut, and if Vox's "2.0" era fails then I'm not sure what he'd do at that point but leave the company since he's sick of being pidgeonholed as a walking dildo.

Other than that the only other big talent I can see gearing up to leave is Selen. Fat jokes aside, it just feels like she's been out of it for a while now, not just due to APEX but with that contest where fans design her a new costume. As an artist she was probably pissed off with Anycolor's original T&C, which basically said that even if an applicant doesn't win they can take their design and repurpose it in any way the company sees fit without any form of compensation. The fact that she probably had to shell out $5k USD of her own money to 5 lucky winners and that "Tired" tweet she did on her PL immediately after makes it look like that was the breaking point.

That's debatable. Yes Pomu is a Niji fan, but its clear that she doesn't enjoy working for Nijisanji anymore and is only staying on for her fans/friends. She likely renewed her contract so she probably won't leave this year, but unless things really change up and she finally gets Home3D early next year, I don't see her staying on for long.

As for who will graduate next? The candidates:
Enna: Keeps doomposting about graduation and talking about it constantly, apparently had gotten in trouble multiple times with management. Will probably not leave willingly because she's addicted to money.
Millie: Depression posting and visiting a therapist for her mental health issues caused by her job. Still, she needs money and won't leave voluntarily.
Reimu: There are theories that she's very rich and has no reason to stay. According to her she has rich family members but they don't give her much, so she has to pay for things herself. Probably not leaving voluntarily.
Vox: Has gone into a full blown mental breakdown. His numbers have collapsed and he's changing his content too. Although he's made enough money already, he clearly wants more.
Shu: The most likely to leave. Long rant about his deteriorating mental state after the AR live was shut down. Took months off streaming and seems to hate working for Nijisanji. The company wants to keep him just so they can keep selling Luxiem merch packs, but the merch doesn't benefit him anyway, so if he wants to agree to be their paypiggy bitch that's up to him.
Pomu: Everyone wishes she was in Hololive where she belongs. She's good friends with Kiara, and her friend Nina is leaving. As an indie she struggled a lot and she probably doesn't want to experience that again, but if she got an outside offer she might just take it.
Rosemi: The slowest growing channel for a year now. The only seiso vtuber who sticks out like a sore thumb. NijiEN is filled with rudetubers who drive away her potential audience. And she's abandoned companies before.
Kyo: Doesn't really belong as a vtuber, he's a much better fit for twitch streaming. Kind of puzzling why he's even here. But if he graduates it will be next year when his contract runs out.
Assorted low-view garbage (Ren, Ver, Aster, Doppio, Aia, Kotoka....can't even remember them all)
They might graduate when their contracts are up simply because they're low-viewed garbage on a sinking ship. Nobody really cares about these guys though.
