Some versions are harder than others, and it also depends on regions. In terms of difficulty though what matters the most is whether or not the version you're playing as auto-aiming.

In terms of content, director's cut has one "extra" scenario which changes enemies/item placement (including key items) and changes other bells and whistles too.

Saturn version has a super fun unlock-able Battle Mode, the best costumes (imo) and a new skin for one of the enemies along the way. Saturn version has no auto-aim in any region AFAIK.

The DS version has another "director's cut like" extra mode but it's god awful, it adds anti-survival horror forced first person knife fights and replaces puzzle by touchscreen shit that's on the level of a toddler's educational toys.
But it also changes content for the "classic" version of the game: all 3D models were redone from scratch, characters and enemies are more detailed. The player is given two more moves: 180° turn and RE4 knife-use (you can take it out with a shoulder button and it doesn't take place in the inventory). To make up for that, zombies have a smarter AI and are harder to dodge.

PC has no extra content besides two shitty guns (which both play the same are basically rapid fire berettas), but of the classic one it looks the best and it has modding support.

There is "definitive" versions and they all have their pros&cons. Go for Director's Cut (non dualshock) if you want auto-aim and want the most content. Go for Saturn if you want no auto-aim and the battle mode. Go for DS if you want a more modern way to play the game.
