Depending on if the graphics file has transparancy stuff built in you may or may not need to fiddle with the existing w/threshold / transparant color cyan radio buttons. PNGs usually have transparancy built in so we can probably ignore that part though. Click on the dropdown box under the image on the right and move it to the DOOM color palette, then click convert.
