We all know the finger on straw works but I was wondering if I placed a stopper on top of a tube that had several honeycombs and put water in each layer would the water stay?

No, because water can flow down the tubes on one side, and up the tubes on the other.

Whichever tubes have slightly more water in them will flow down, and the ones with slightly less will flow up.

And by tubes I mean the little honeycombs.

You mean if there was a flat surface beneath the bottom, and air couldn't get in around the sides? That would keep water from flowing out of the bottom, but it wouldn't stop water from flowing through the two honeycombs in the middle. I don't think that was the OP's intention, though. I think he wants to have the bottom hanging in the air.

it doesn't matter if some water drips to the bottom as long as it does loose all of the water in the system, the plan is to have about 15 high of honeycombs, as long as the pressure will reduce the flow of the water downward enough to make it viable to use.

seems obvious, the honey comb would make zero difference to the pressure, so whatever level the water was at when you put your finger on it it would stay at, just like the empty straw.
