What's next? Commercial crew to the ISS sometime around 2014-2015, assuming all goes well. SpaceX, Boeing, Sierra Nevada, and Blue Origin are in the race for providing crew transport services to the ISS, and presumably at least two of them will be selected to fly to the ISS.

Beyond that? Who knows. As of right now, NASA has no definitive plans for exploration beyond low Earth orbit. Obama directed them to visit an asteroid by 2025 and go to Mars maybe sometime in the 2030's, but as far as plans go, that's as weak as it can get. No definitive launch vehicle design has been selected as of yet, no payloads have been confirmed, and the only thing that's been confirmed is that the crew will be riding in the Orion capsule from the Constellation program, which is relatively close to completion.

Hopefully NASA headquarters will stop dragging their feet and actually announce something soon.
