Yes, mostly on mice.

I break their necks, cut them open, take out their spleens, grind the spleen. From the bloody suspension the B-cells are isolated which are then infected with recombinant retroviruses to deliver a genetic payload in a specific genetic background. Say the mouse is gene X knockout I use the retrovirus to deliver a variant of gene X (some mutant or other) and study how this variant impacts B-cell response to a stimulus.

To be honest I didn't enjoy this shit one bit.
