Abstract

We tested the effects of Fe(NO ) on the growth, DNA expression, and RNAi pathways of Arabidopsis in wild and mutant (DCL knockout) types. DCL genes in Arabidopsis code for part of the RNAi system. Iron Nitrate is found regularly in soil; it is a trace nutrient for plants. However, this nutrient has a very high potential for biotoxicity in higher concentrations. The purpose of this experiment was to find differences between how the wild type Arabidopsis responds to Iron Nitrate stress and compare it to how mutant Arabidopsis responds to Iron Nitrate stress to determine specifically how DCL genes play a role in the RNAi system when responding to Iron Nitrate stress. We used Arabidopsis Thaliana because it is a genetic model organism. Observations show that DCL gene knockouts tend to grow much slower and have a shorter life span when stressed with Iron Nitrate. In other words, the plants without the genes to code for the RNAi system were observed to have greater difficulties growing and staying alive than plants with a full RNAi system. Stress measurements that were taken included: time to reach developmental stages, leaf number, bolting, coloration, and root growth. Some plants were even observed to have lost the ability to undergo photosynthesis. This tells us that the RNAi pathways in Arabidopsis have a relation to regulating homeostasis with regards to Iron Nitrate in the plant.
