Im means that we should acknowledge that our desire and our dreams as well as our common social imagination is structured by fictions. Right now we are pretending, that everything is as it is and has to be done like it is being done because thats just the nature of reality. However human reality and the way we deal with it is totally controlled by fictions or virtualities like language, realigion or even narratives like science or forms of government. we pretend like all of this can be rationally deduced and deny that in the end we made it all up. to take fictions seriously means to reflect those fictions and become aware of their genesis and implications and ultimately to transform them to form a more ceherent, just and meaningful future

He's talking about the concept of Hyperreality, which Baudrillard coined and analysed. Simply, it's the idea that reality and non-reality are now indistinguishable.

Read 'Simulacra and Simulation'; Zizek is pretty much just building on those ideas anyway. Also read Debord and Eco, etc.
