It's sort of a convenient slogan for his entire philosophy. You can trace almost everything Nietzsche said back to "god is dead" in some way:

1. The decline of religion has left a void in our souls.

2. The coming of nihilism.

3. The need for man to aspire to god-like status.

4. The decline of "moral certainties."

5. The urgent need to establish some kind of secular god (ubermensch).

He means that the Christian God is no longer what guides people to do this or that and so he proceeds in talking of all of the substitutes for God and his own thought on what we can do in this scenario.

God is one of the many metaphysical excuses, that which commands and looks over you, that protects you and guides you. Nietzsche knew how easy it was to be guided, if not for God, for something else.

It's not a "God does not exist" quote as some atheists put it, even though that doesn't mean to say Nietzsche believed in God. If God is dead is only because he was alive. But here, again, to be alive is to exert one's power and if you believe in God and follows his way, that's it.
