That's a bit of a loaded question. Most, or at least many, western philosophers agree the most important thing is to be happy. However what if your happiness involves inflicting pain upon others?
I suppose that'd lead into your second question. I consider some of our responsibilities to be being kind to one another, tolerance of culture, not eating animals [inb4 hur dur preachy vegan], and most importantly, at the end of the day, being happy. Of course that 'happiness' should not be 'pleasure' as you might get from inflicting pain on a person, but real, true, happiness.
