To answers OP's question, I would say no. It is not wise to start with a book which you can call a "Magnum Opus". Something like that is usually a rather extensive project, and it is unlikely that anyone will publish an 800 pages book by someone who has never published anything before.
It would probably be better to establish yourself as an author, and then take the time to write what can be called a Magnum Opus.

There is a very good example of a guy here in Brazil that didn't get published (Eduardo Spohr). He did it himself and invested heavily in marketing. Of course he had friends that were kind of famous in the internet scene.

His books are completely shit, though. So it was obvious why he didn't get published at first, but now he is a nerd celebrity.

You can go more casually, because some people like GG Marquez just decided to write full time before actually being published. He could have started slowly, as a second job, get published for the first time, see how things go and then jump in (more like bukowski, maybe)

There is a difference between a masterpiece and a Magnum Opus.
For example. Portrait of The Artist as a Young Man is a masterpiece. Ulysses is a Magnum Opus.
The same with White Noise / Underworld.
