Depends on what you're going for. If the greatest book ever written happened to be a fictional biography about Icelandic singer Bjork's hardships during WWI, I wouldn't think it was all that great, as I am generally disinterested with that subject matter.

So yeah, there's a difference between best & favorite book, but you have to really like a book to say it's the best.
