If you consider La Celestina as medieval, it is. Also of that period Prison of Love by Diego de San Pedro is interesting, it's in the Bloom's canon.
But the best medieval Spanish literature is in valencian/catalan.
Tirant lo Blanc and Curial e Güelfa my favourite. Also in catalan there's an interesting unique author, Ramon Llull.

Also the medieval old 'Romancero', recopilations made poems called 'romances'
The 16th century admired these compositions and did not hesitate to imitate and revitalize them. Authors such as Lope de Vega, Góngora or Quevedo wrote romances in the manner of the ancients, forming what is now known as the New Romancero.

I'd add a really good one from medieval Valencia: "Llibre de les dones" (the Women's book) by Jaume Roig. It's basically a compendium of poems with the coolest metrics ever that rants about how horrible women are. Also there are really cool little stories there, like one about canibalism

Don Juan Manuel's El conde Lucanor. El Amadís is not only really fun but also very influential. Las coplas a la muerte de su padre are essential and is a very beautiful work.
Some writers or works with which I'm not as well familiared but which I've been told to be important are Juan de Mena's poetry and specially Laberinto de fortuna, which if I remember correctly follows the model of The Divine Comedy; the Marqués de Santillana, who, if I remember correctly, has some poetry similar to the one you'd find in El libro del buen amor, is considered as a good poet and was also who introduced the sonnet in Spain, although his were very bad. There's other stuff like galaicoportuguese poetry, the jarchas and the dance of the death but those are not very interesting to most people.
