I haven't read the Bible and I did come out of the Brothers Karamazov feeling like I hadn't understood very much, but I enjoyed it nonetheless.

You can understand much of it without knowing the Bible well, but your experience will be enriched the more you know about Christianity and Scripture. It's the same with anything- you'll enjoy the book a lot more if you're familiar with Shakespeare and Gogol and Pushkin, too
