How do I measure the cost of making a book to self publish?

So you are not asking about the cost of printing, but rather what to bill for the design work etc you do (on your own book)?
Just try to estimate how many hours it will take, then either take an arbitrary hourly rate (maybe equal to your day job) or research market pay for this kind of work and multiply by number of hours.
