It has to do with your beliefs and values. Essentially the superman "lives by their own religion" overcome the any belief in an afterlife, and live by and enforce one's values with a spirit of self-confidence and nobility. Similar to how Zarathustra, the legendary founder of monotheism is.

Also the concept of the superhuman clashes with the concept of the Last Man. Wretched nihilists who are sort of like some of the people who browse this site, who are too cowardly or animalistic to fight for their values from within and have it be their world. Instead they retreat into slave morality, apathy and resentment.

Nietzsche saw a process in western civilization that would lead it to this point which in large part captures the contemporary cultural Zeitgeist.

Although Nietzsche's philosophy does point out the existence of natural inequalities, it has nothing to do with biological difference per se. It does have a difference in some innate source of power, motive capability, self-assertion. So this notion of a "will to power" is what is supposed to set superhumans apart from last humans.
