Infinite Jest

I think Infinite Jest. I’d say there are actually better American novels, like Blood Meridian, but Infinite Jest focused so strongly on problems that plague modern day America I’d say that makes it American.
I’m also going to be a shill and recommend the Great American Nove Podcast hosted by Scott Yarbrough and Kirk Curnutt. They’ve discussed Moby Dick, The Great Gatsby, and Huck Finn amonst others.

Mario Incandenza from Infinite Jest has that Mister Rogers feel.

Mario Incandenza from Infinite Jest gives the Alyosha vibe. There's a great scene where he touches the hand of a homeless guy and restores his faith in humanity. It's at the very end, but it's worth the thousand pages.
Also, early in Les Miserables is Bishop Myriel. He doesn't show up a whole lot though.
