There have been several,
"Of Human Bondage" by W. Somerset Maugham, it's hard to explain but you only really "get" this book if you've had a hard, lonely, poverty stricken youth like i have and like the main character in the book. It made me feel less alone in the world and made me realise the real importance of taking my own life in my own hands, being responsible for my own happiness.
The passage where he is in the British Museum looking at ancient Greek tombstones and wondering what the point of life is, this is one of those passages i want to read again before i die, perhaps even have it read at my funeral. It is profound and beautiful.

I was just going through my 2021 reading list on Goodreads.
It turns out that i didn't read a single book by a living author all year.
Dead white males with the exception of a couple dead white females.

Two big ones i can think of

"Of Human Bondage" by W. Somerset Maugham, made me realise my own hard upbringing and young years full of loneliness and poverty were not an isolated thing. That other people had survived and overcome similar miserable starts to life. That life could still be meaningful, worthwhile and beautiful even after a youth full of misery.

"The Forsythe Saga" by John Galsworthy. Made me think about what life is really for. The whole basis of what one lives for.

I can't explain it better than that.

"Of Human Bondage" by W. Somerset Maugham is the original "life affirming" book and i don't mean that in a cringey way.
Based on the author's own life it tells the story of a young man's struggle to find his place in the world. He experiences poverty, loneliness and dissapointments but eventually gets his "sort of" happy ever after.
You should read it.
