Sometimes, not always. It's like a primitive, animal-like way of saying "I'm happy, and I also trust you're not going to rip my guts out, so I can close my eyes safely around you." It shows you trust whoever is around. So it makes the smile more special, even if the other person doesn't consciously realize it.

It doesn't represent eyes being closed. In fact, it's impossible to really close your eyes and end up with the lines where your eyelids meet looking like upward curves. However, when you smile (like a real, genuine smile) your lower eyelids do get pressed up, causing less of the eyeball to still be exposed. If they get pressed up enough, then your eyelashes will cover up the entire part of the eyeballs that's still exposed, making them kind of look like thick black lines. This is also why some artists draw little white blobs on the eyelashes when drawing a smile, representing the parts of the eyeballs that still shine through the eyelashes.
