Ayako's route I always took as a diverge from when rin erase's your memory in the start of ubw and you get the normal/bad end that isn't a dead end.

You and her hit it off and then her and you run for your fucking lives when a giant brown guy with an albino loli on its shoulder is trying to kill you.

Seek sanctuary at the temple, caster gets involved or shinji rescues you because he was always tsundere for both Shirou and Ayako.(then rider takes a piece out of both of you instead of just Ayako awwright).

I always saw Ayako as Shirou's path to a normal life.
And as sad as it may be, I also got the impression that Ayako could possibly have had a slightly easier time to understand Shirou (Rin's mindreading of Archer disregarded).
All of of the heroines came to understand how Shirou had grown to become incredibly stubborn, and how he was broken somehow compared to a healthy mind. But I believe that Ayako could have possibly gotten some insight as to how this form of thought came to be. Heck, despite all of the magic and fighting of which the others have more in common with him, Shirou did all these things the same way he lived his life, and the same way he saw his Archery, and these things she shared with Shirou, or at least had insight into, which the others didn't on the same level.

But I could have still found such a route to be incredibly interesting.
Say the war started somehow, and something unexpected happened and Ayako got involved, an extra, an outsider, a normal factor having stumbled upon their abnormal event.
Nothing like it happened in the other routes, the closest thing would have been Shirou himself having gotten involved. Which also brings on another point as to how they would relate.

Despite all of the abnormal in Shirou's life, he had still lived a "normal" life with slight knowledge and practise of the abnormal, rather than the other way around.
So yes, I believe an Ayako route could have easily become something of the most interesting in the novel.
She could in a way double as some kind of anchor to the normal side of life, while at the same time she could relate to Shirou's abnormalities and he to her normalities.
It could have been amazing.

The point of HF was that Sakura was Shirou's path to a normal life and vice versa. An Ayako route would be interesting, but it's kind of hard to make her fit into the plot. I could see her as Archer's (who might have been someone completely different, like Robin Hood) Master back when Rin was supposed to have Lancer, though.

HF ends with Shirou bursting through all the abnormalities and shatters everything to hopefully end with what an ambition of how things should be.
Ayako already lives in a way of how things should be, a normal life. She could either flow more into the magical side of the world, or she could serve as a sort of a counter weight and prevent Shirou from becoming further sucked in to a certain extent. Even if they were to meet somewhere in the middle which might be the most plausible, it would still be more normal for Shirou.
