Considering the "main" playable characters, you'd probably have Sanae as imouto-type who looks up to Reimu and tries her best; Sakuya as the cool older sister who acts like an enemy but helps Reimu out because "we just happen to share a common enemy".
Marisa is of course Reimu's eternal (self-professed) rival who just can't win against Reimu, but she'll help Reimu against strong opponents because she likes to fight and because she won't let Reimu lose to anyone but her.

I'd love to add Youmu but taking care of Yuyuko is a full-time job.

Cirno is the recurring comedic midboss. "I'm the strongest in Gensokyo! This time I will defeat you!" The trick is you never actually fight her, as she always manages to defeat herself. For instance, creating a huge block of ice preparing to drop it on the group from high up, only to get squished by it. Or Yukari gapping in some item that the team needs just in the nick of time, right on top of Cirno's head.
