1. Her name
2. The Luna Dial
3. Her appearance
4. Her ability's similarity to Kaguya's
5. That ZUN explicitly suggests in her profile that she's likely to be much older than she actually appears while still being human.

That said, I don't think they're sisters - isn't Eirin essentially supposed to be the ZUN version of a particularly Shinto God? It would be more likely that Sakuya is a descendant or distant relative, ala Toyohime/Yorihime.

While it's never stated that they are the same person, Eirin is identical in description, function and part of her name to Omoikane / Omohikane no Yagokoro. Ie, she predates Amaterasu, Susano, Inari and the whole gang of popular gods, including Tsukiyomi himself.
