To answer that question, you must first understand Mokou. Fujiwara no Mokou.
For starters, Mokoutan is a loner. Due to her childhood, Mokou is a very lonely girl of very few words and so little is known about her. Confessing your love to Mokou might not do anything since the only times you will normally see her is with either Keine or with one of the heros of Gensokyo (incident starters or solvers) if not her acclaimed Yakitori stand. All of which you might not have a lot to say or converse with her over.
Mokou might have a few things to say but don't count on being buddy buddy with her. If you get lost there or are escorted through the Bamboo Forest of the Lost, then that is possibly your first, only and last chance of being alone with Mokou. Given her nature, Mokou really wouldn't care all that much, but she's still a girl. She might even be inclined to avoid you from that point onward. Unless you're someone even a tad bit more valuable than the normal human villager through super special means, or someone noteworthy like Kosuzu or even a friend of Keine (hah), you aren't going to have much luck with her because not only is she quiet and rather secretive, but she has also been around longer than you and will more than likely outlive you. Intimate feelings are probably not foreign to her and probably had one or two every other decade confess to her. You probably aren't the first but you also aren't the last. Unless you're a cut above the average human villager, you probably won't see eachother often. The next option is to be friends with Reimu and decent enough at danmaku to defend yourself and a high enough alcohol tolerance to not get wasted too soon.
She'll show up at one of the events or parties so just be patient and hope she shows up so you can speak more and get to know eachother, eventually become friends and have a meeting place or signal for whenever you two want to meet and go do something for a change of pace. You'll be good friends with Mokou but I doubt you'll be able to court her. Even if you do, it all really falls back on if the love is genuine and if Eirin is willing to make another batch of Hourai Elixir. Mokou is lively with those she sees as friends but getting through that cold/quiet and secretive exterior of hers is probably the hardest challenge if you want to get to know her and be close friends.

Word of advice, don't get manipulated by Kaguya-don't be buddy-buddy with Lunarians. Keep it formal and nothing else. Not even paizuri from Lunarians. Don't even dare look at them for more than a second when passing by. Even if they're incredibly attractive and catch your eye for more than a moment at a time.
