How do I play this old 1995 game? Not Touhou.

I downloaded the CD and ran set up, then mount the CD on a virtual drive (like E:). I run the translation .exe with LocaleEmulator in Japanese, and everything launches but says it can't find the CD. I've tried different drives, but that's it.

The virtual drive program I use is WinCDEmu, is it just the program and I should try a different one? Or should I burn the CD to a physical disc?

The virtual drive has nothing to do with it.
The problem is likely Windows.
Older games simply don't run on newer versions of Windows, or if they do, they run with caveats. Like crashing, or running in "turbo" mode (which is what that similarly named button on your PC is designed to prevent)
Try running it on a Windows XP VM with AppLocale installed.
If not, try Windows 2000. Then Windows 1995.

HyperV, ESXi, Xen, VBox, VMWare, whatever.
