So, they isolated Shiki's DNA from the scene and they were able to conclude from the sheer lack of anything else relating to Shiki (severed limbs and such) that he was a suspect.

How do they know that Shiki was the owner of the DNA?

I'm sure somebody saw him enter the hotel and possibly leave so it's not out of the question they'd figure out it was his.

It's easy to tell because it'll have lots of sounds that sound like voiced inhalations instead of exhalations. Some languages have that, but those are rather rare and not used much in those languages either because it's harder to do.

When you just play a tape of someone speaking backwards, it ends up sounding like half of it is inhalations. I remember the foreign dudes in Wings of Honniamise had that, and a few other animes have had it, I think.
