Mokou is deeply introverted. That's why she lives alone in the woods and she won't even talk to the people she rescues from Youkai.

But somehow she seems to have developed a close friendship with Keine.

I would assume that, after centuries of losing everyone she cared about, Mokou tried to prevent herself from forming relationships. That's why she became so introverted. But somehow she ended up befriending Keine, even after trying to force herself away from other people.

I think that would make the loss of Keine especially bitter.
