We know that Shiki can't kill Servants permanently, but do the lines appear on the Servants so they can be killed while alive during the holy grail wars? Wouldn't that also kill the mage?

It wouldn't kill EVERYONE she knew. Hisui would survive, and so would doctor Jinan. She was sort of hoping that Shiki would emerge the victor, as well, meaning that the only people to really die would be SHIKI and Akiha.
