Long story short, it's a series of poor quality Touhou voice dramas that were published in early 2010s in Nico Nico (don't know if they have made more). The series got a shitpost reputation for being so bad, and was associated with Japanese amateur gay porn, which was also shitposted on the site.
The director of the series got mad for the backslash and tried to get all the Cookie parody videos deleted. Nico Nico users digged into this director and found out he had used the series as a way to "get to know the voice actresses better". Later the director would kneel before Nico Nico users and give them the right to remix the shit out of Cookie.
If you're wondering about all the several Reimus and Marisas, they're separated from each other by their voice actress. One "type" of Reimu is voiced by one voice actress. The other characters that occur are named after their voice actress, separating them from Touhou overal

This sums it up pretty well, but I think it's essential to detail the Manatsu no Yo no Inmu connection given how much that community crosses over with and has fundamentally influenced Cookie from the beginning and made it what it is. The sheer volume of parodies and otoMADs, the extensive tag 'genres' thereof, acronymisation of the actors' names, direction of the later dramas etc. are all direct results of the 'homo' community latching onto Cookie due to its commonalities with Inmu (bad acting/bouyomi, melodrama, generally poor production, etc) after the tag vandalisation of the original video. It's a big part of why Cookie is considered to be a mostly separate entity to Touhou rather than just another derivative work - the constitution of the culture that shaped it and exists around it is very different.

The actors are significant as well given that so many of the injokes and characterisation of the 'characters' in parody material and the later dramas is derived from them, their online presences, personal idosyncrasies and various bits of real life information dug up by elements of the community, as well as forming the moral quandary that is probably the barrier to entry for a lot of people.
