Forte gets a huge buff in the second. She's still slow as shit, but at least she actually does a lot more damage compared to the others. The flip side is that Chitose is pretty damn broken. She does slightly less damage than Ranpha's anchor claw per shot, but gets about 5 and will switch targets to any in the nearby area.

The routes diverge about halfway through the game. At that point, you get to choose between the three girls you've scored the most "points" with. So if you keep a savegame there, you can play three routes from there, then restart the game from the beginning to go for the remaining two routes.

There's also some stuff you can discover about Prince Shiva that slightly affects the story of the second half of the game, and the ending. The Prince Shiva stuff is completely orthogonal to the five routes. You can do it in your second playthrough, or deliberately not do it in your second playthrough if you managed to discover it in your first.
