Stage plays are acted, movie scenes don't occur in real life, magic shows have their secrets, and animation doesn't occur in our material reality Any show that we like presses our "moe buttons," and it is our individual preferences that causes us not liking the same sets of shows.

Virtual Youtubers are kinda like idols, they're real people who put on a persona for the sake of entertainment, a fabricated character. Only that this character is also a 3D character in a virtual environment, hence the name.

Voiceroid let's plays do not enter this category, because while the creator takes care to roleplay them as characters, it's more of a manzai or a sketch comedy that's tangentially related to whatever theyre doing or playing, there's no relationship with the creator who simply writes the gags, while a virtual youtuber has to act them out and come up with stuff on the fly.

Not to shit on voiceroids thoughI actually like and prefer them over virtual youtubers, they have some amazing gags and creative stuff they've done like making them sing (despite not being designed for it), an entire subculture dedicated to them, their own yukkuri-like things, commentaries on all kinds of stuff, etc etc, but fundamentally they're just different and seperate from Virtual Youtubers.
