They probably would have used other heros or just left the heros (except for Archer) and change the genders of the main characters (Shirou and Saber).
Think of it. (Female) Rin sexes up Saber and introduces poor Shirou into sex. Shirou, who is a virgin, isn't too eager at the beginning, but decides to overcome her doubts for the man who saved her once and almost died by doing so.

Only other characters which would have to change would be Sakura and Shinji. Mainly because Sakura is in love with Shirou but is unable to confess (and Shinji is making his life hell). Also, incestous love between Sakura and her lost big sister Rin.
