Fanon works often uses her humanity as a base for romance drama, which I always found really odd because I can't remember her showing any opposition to the idea of becoming a youkai.

Sure, she is eccentric and sometimes weird, but she is ordinary in the sense that there is nothing inherently special about her and her origins, kinda like you and me. She was born an ordinary human and worked hard to get where she is today, meanwhile Reimu was born with her abilities.

Sure, she could become a Youkai and use magic a lot easier, but Marisa would rather get there through hard work and guts and become a powerful magic user in spite of her normal human side.

Well it was mentioned that she was trying to create an immortality elixir in several games ,but failed in the end.

There's really no downsides to her becoming a youkai. Marisa knows this, she hangs out with youkai all the time and can tell that, the same as people, there are youkai that are bad and youkai that are good. There is no reason why she wouldn't become a youkai. I don't think it would necessarily be a conscious ritual or decision of whatever. As she continues to accumulate power, at some point she is no longer human and is considered a youkai.
And I don't think Marisa would ever stop studying and strengthening her magic so it's pretty much inevitable that she will become a youkai

In PCBs manual, it was mentioned that she does make attempts to become immortal. Also, in DDC, she's the only one of the protagonists to be mistaken for a youkai by Seija; the other two are recognized immediately as "just" human.

There's two stages to it. Stage 1 makes you a youkai who doesn't need to eat. Stage 2 is optional and stops the aging process. Patchy was born at stage 1, but any human could reach it if they figured out the right spells/potions.
