I'd have to say Koishi, because she actually lost something, even if she's not particularly unhappy about it now.

Stories of all the other youkai might have started up sad, but in the end they all ended up with cool abilities, indestructible bodies and (most of them) lots of friends, so it's okay, all's well that ends well.

It's post-elixir consumption where things become dark as fuck, as for 600 years after, she is either hiding in isolation or killing anything and anyone she sees. Logic indicates at the time she had completely lost her grip on sanity.

Not really, every man that tried to marry her had to complete the 5 impossible requests.

Kaugya isn't that of a bitch, at first she was, like every Lunarian that thinks about the Earth being a place for losers, then thanks to human contact she gets friendly and more open.
Also at the end Mokou doesn't really hate Kaguya anymore, just read the part where she learns about the possibility of Kaguya returning home, she was a bit sad and tried to confirm things about that.

Although hers might not be the saddest, I always thought Tojiko probably had a pretty shit life. She was betrayed/used by everyone she trusted and was locked in a hole for 1400 years.

No wonder she became and evil spirit. She might have done her fair share of bad deeds in life though, since the Soga clan weren't exactly the cuddliest, killing other clan members like it's no big deal and all.

Did you listen to Shikieiki, the cycle repeats because of Gensokyo's traits. The 3 heavenly lights, the 4 seasons and the 5 elements multiplied together are 60. Shikieiki is quite determined with this, which leaves us the implications that Gensokyo may have required, or caused, or perhaps done both, as the second world war. Another 'catastrophe' that we lack details for happens during the events of PoFV. Can Gensokyo just suck up the dead of any current catastrophe, or does it cause it, we can't tell?

Not that anon, but in the Tale of the Bamboo Cutter, the Jeweled Branch of Hourai really was a fake, and that he really did try to trick Kaguya-hime. She believed him wholeheartedly until three workmen burst in and demanded payment for the jeweled branch they had worked on for so long, and so he went away in shame. Here's one version, though it has no specific names; every suitor is just referred to as a prince. I'm sure that there are other, more complete works online.
