The Nanayas hunted nonhumans, specifically ones who went berserk like the inversion principle of the Oni clans.
Their Pure Eyes basically allow them to "see" mystery, but they themselves have almost none. This allows them to be perfect anti-Mystery assassins because while their marks see them as normal non-mage humans that aren't part of the masquerade. So then they move in close as a normal human would and kill said mark.
Shiki was able to walk right up into Arc's home and cut her to pieces before she could react, for instance.
