Are you talking about vocaloid in general or just Miku? Vocaloid allows for some very low barrier of entry in making songs, anyone with a computer, some effort and some talent can create something, and you have already an audience thanks to the brand, which allowed for some pretty creative and diverse set of works.

Miku is the most popular probably because she was the first (more) anime style Vocaloid on a better and easier to use software (Vocaloid 2). It's possible that any other Vocaloid in the same situation would be the Miku (and Crypton was competent in giving her some strong marketing push once she was getting popular). And once people loved her, any attempt to make her less robotic would inevitably not catch on (like append).
