Outside of his words, you can see it by how he draws her eyes. Eva is the one drawn with Takano's eyes (with the long eyelashes even bellow her eye), not Lambda. So, he definitely could have made her face more Takano-like even in Umineko's art style. Not saying her eyes specifically resemble Satoko's either though, they're just a more generic shape, compared to Takano's (and Eva's) more unique ones.

Art that makes Lambda's face look like Takano's (like Pachinko's sprites and Umi manga episode 2) always felt really off to me.

Now granted, aside from the pie puzzle, none of that would point specifically to Satoko, and the puzzle is obviously something very minor, but my point here is more about showing that Ryukishi's claim that Takano isn't Lambda has some hints in the work itself, in spite of her apparent similarities with her.

Lambda's hairstyle obviously mimics Takano's, but look at her eyes. They just aren't drawn by Ryukishi like how he draws Takano's eyes, where he highlights eyelashes on top and bottom of her eyes. Rather than that, she has a line going from the top to the external side of her face rather than just top and bottom.

This eye shape is kind of generic, so once again, it's not really Satoko specific, but the main point is that it doesn't line up with Takano's face just like how he claimed Lambda doesn't look like Takano in spite of the colors and hair style.
