Shiki could probably understand "Servants" as a concept of "Familiars", and would be able to Kill them.
However, he could not Kill their heroic spirits and they would be resummoned by the Holy Grail.

Of note, however, Shiki could not hope to match up to a Servant physically, and his head would fucking pop if he tried to Kill their Noble Phantasm.
