The healing of injuries caused by the elixir can take up to days, it's not Wolverine-tier so they don't necessarily restore their copious amounts of milk instantly, that said it is pretty clear they will regain their amounts faster than regular humans which means they'll be able to produce more than normal.There's also the "age" of their bodies to take into question, since Eirin has a mature body she will be able to produce more milk than Mokou and Kaguya.

However, they still feel pain and exhaustion like any regular person, so extracting 96oz of milk will be as tiring to them as it would a normal human woman.
