Caring if Yasu was born a female misses the entire point of her character. She was obviously born male as seen with the man form 19 years ago and Lion but it doesn't matter since Yasu is Yasu not whatever gender role you assign her. Her entire character is about her struggle in accepting what role she want's to fit into which she finally solves and accepts herself in episode 6 by choosing her female persona so yes Yasu is female even if she was born with male genitalia
