In E+ weapon and armor weight affect directly your attack and defense. In vanilla flying scrolls directly increase/decrease equipment stats while also modifying weight, but the weight itself does not directly make items stronger or weaker.

My understanding was that equipment weight in E+ only affected your crits and crit resistance. I tested it out in wizard mode, adding about 30s to both a weapon and a helmet. It looks like weight does have a small direct effect on regular attacks but not defense.
