Not really.
Making Comics by McCloud has some "rules", but they're really basic, like these:
Avoid tangents.

Stick to one action for panel.
Follow the 180 degree rule.

The point is to make the reading as easy as possible and to avoid confusion. As long as you do it, you can do anything you want.

Then American Comics, Bande dessinèe and Manga have each their own "traditional" paneling, but the best way to learn it, it's to read comics.

Bande Dessinèe for example are slightly bigger than comics, so traditionally they have more panels for page and there is a lot of space for backgrounds.

2x2 format is SHIT for anything narrative. I have tried it. It's shit. If you want to see how bad it is check the comic Endtown, which is a competent comic but completely crippled by the awful format.
2x2 is only good for comedy strips or 4koma.
I don't know instagram but I would rather make a sequence of square images, or even better, host the comic on an actual fucking website and post a frame or something nice to look at in the instagram, with a link to the comic in your profile.

Wrightson didn't use his brushes too much for the Frankenstein pieces, they were made with Gillote 303s I think. I guess it's just the subject matter and the background hatching that makes me connect the artist with Wrightson.

I started Made in Abyss yesterday, it's really pretty and unique. One thing it does that I haven't seen elsewhere are these first-person pov panels.
These highlight small details in the setting that I think would be difficult to show off with more traditional panel layouts (curved beams in the ceiling, cracks and trinkets by the mirror). If you spend a lot of time thinking about environments it might be worth trying this.

I'm still at the beginning but so far it's been pretty good. It has some really fucking hype moments, made even more hype due to the art. It does kinda meander a bit at the start, but once stuff is set up, it's quite good.

The serious/silly thing just seems to be how it is. I actually don't mind it too much, gives the manga some heart. You grow to like the characters a lot. Plus it doesn't take itself too seriously and come off as pretentious or 2deep.

You're already doing a ton of Japanese things for no practical purpose so why not. You can decide on a case by case basis whether it's necessary for the reader to know what the characters mean. Translated manga where the onomatopoeia are left as is are usually perfectly readable, though it's helpful if they have a tiny footnote at the bottom of the panel with translation
