Two common ingredients of comedy are the unexpected, and the boundaries of social acceptability.

Drawing a dick on the school whiteboard is both these things to teenaged kids. Drawing a dick on your passed out drunk friend's face is both these things to a college aged kid. Etc.

Drawing a pair of boobs is just about equally common for the same reasons, but usually also done by men. The reason women don't usually do this (and the reason the stereotype of women not being funny exists), is because they have no need to be.
Humour is a sign of intelligence and being social, because it requires an understanding of your environment to know what's unexpected and on the boundary of social acceptability in a funny way. That's why many women cite "humour" as one of the most important character traits in a potential partner.
Men rarely cite this. Men have other preferences in female partners, thus women don't have a need to be funny, and that's why they don't draw vaginas on things.
