Hi, Basically i sketch and do line on 1 layer. Flat color and render on another layer (i still use different layers for different part like hair, outfit, background...).
Occasionally if i'm not so sure about level i'll add a multiply or overlay layer to adjust the shadow, or a hard light layer for highlight, reflect light, ambient light...then merge to the main color layer right after.
