It's done by an "organoleptic" test, which is a fancy word for a fucking taste test. They just have a panel of judges either taste a solution made with the peppers over and over again at decreasing strength until half the judges say they can't taste it any more. It's hilariously poorly thought out.

They can use liquid chromatography to measure capsacin density, but all they then do is compare it to previously rated peppers using the highly subjective and flawed scoville scale.

mezcal is one of the most diverse spirit categories i've ever encountered, and it's really difficult to tell what's what even if you're really experienced with it. i've had a mezcal that tasted like a gin with some creamy notes. i've had a mezcal that tasted like a bell pepper and made you want to vomit. i'm currently drinking a pechuga that has just the right amount of smoke -- just a little less than a non-islay scotch. The fact that scoville units don't map linearly to pungency units is comparable to the problem of using colour scales in scientific papers. We're only just starting to come around to the idea that the nonlinearity of the human visual system means we need to design subjectively-homologous scales to display data (look up the design process of the viridis and magma colour scales). Any scale of human perception is inherently subjective, so the goal is to provide an objective characterization and then look at the mappings of that to different subjective experience.
Yes, the scoville scale is shitty and subjective, but that's because it was based on an assumption of linear response to capsaicin. The problem is not with the scale, it's with the misinterpretation that 4,200,000 scoville heat units is 40% hotter than 3,000,000 SHU.
