The details are not fully published yet, since several versions are under testing. However the mana of a node will be strongly connected to its past honest behaviour in verifying value-transactions (proportional to amount of value processed). It will decline over time, so it can not be accumulated infinitely.

How holding tokens goes into that I dont know. However it will not be staking in the sense of "locking" AFAIK.

Mana will be used to have a vote on consensus on the tangle and to have bandwidth in times of congestion. This is the new consensus mechanism IOTA foundation took so long to mathematically proof, peer-review publish and now code for Coordizide.
