He's not trying to profit off of colonizing mars in the immediate future, he's trying to use profits from commercial launches now to invest in future projects that he believes will further humanity.

However once they have 50+ ppl on mars he will be running the only company that has the ability to send more people and supplies to the colony, and no one (especially the government) will simply let the mars colony perish. And boom, guaranteed government subsidies if anything ever goes wrong for spacex.
